Overview

Vanilla

Because of her low defense and low offense, Skye is always taking more damage than she deals out with normal clears. This means she basically starts every match behind. But her high special stat means that she can clear that gap with good item usage. Across a normal round you can expect 3 coins on average, it's important for Skye to make the most of these are they're the main way for her to gain and hold a lead. The items that Skye most wants to prioritize are Hammers, Bandages, and Blindfolds, as those have the highest impact. Heat and Feather are not bad, but benefit the least from Skye's high special stat. Another aspect of having high special is that Skye's self-inflicted items are also boosted, so it's very important to pay attention to which items you are sending when playing Skye.

Flick Instinct

In flick instinct Skye's special stat becomes less remarkable, but her low physical stats remain the same. The benefit is that all her cards are good, which both removes the possibility of hitting yourself and increases the odds of getting a specific desired item.

Strategy

Because Skye gains and widens leads pretty much exclusively through items, she wants to see them as quickly as possible and giving up combo bonus damage to get there isn't a big deal. For this reason it's usually best to avoid setting up chains and instead just go for single line clears as quickly as possible. Once she enough coins to cash them in for an item, Skye really doesn't want the color to get changed on her, so comboing into the item with either a simple bump-spike 2 chain or guaranteeing it with a simultaneous clear are both very common strategies.

Below is a chart for which characters deal more damage in all of Skye's matchups. Note that this chart ignores items and special stats completely, it is not a true matchup chart but a reference for which characters effectively start with a lead in which matchups.
